Combination Adjuvant Platform for Human and Animal Vaccines A novel combination adjuvant platform for human and animal vaccines   Ravendra Garg, Sylvia van Drunen Littel-van den Hurk Volker and Gerdts Abstract Adjuvants are critical component of vaccines. They are being used to enhance and extend the overall immune response, to drive the response towards a specific type of immunity, and to reduce the need for multiple booster immunizations. Here we report the development of a combination adjuvant platform consisting of three immune stimulators, namely host defence peptides, polyphosphazenes and PolyI:C/CpG ODN. The adjuvant platform was co-formulated with a variety of human and animal vaccines and tested in mice, pigs, sheep, koalas, and fish. When co-formulated with a wide range of viral and bacterial antigens including Bordetella pertussis, Respiratory Syncytial Virus (RSV), Chlamydia trachomatis and influenza antigens, a single immunization induced 100-1,000 fold stronger humoral immune responses (IgG, IgG1, IgG2a), a much earlier onset of immunity, a clear shift towards a more balanced or Th-1 type of response and extended duration of immunity of up to two years in some cases. The vacc ines were highly effective in neonates of less than 7 days of age and provided complete protection against a lethal challenge with B. pertussis or RSV. Furthermore, the polyphosphazenes in this combination allow the assembly of microparticles that when lyophilized were stable for several months. Intranasal immunization with these microparticles induced strong mucosal immune response in the upper respiratory tract. Moreover, the adjuvant platform was highly effective in the presence of maternal antibodies. In summary, we developed a novel vaccine platform for neonates, which provided more balanced, long lasting and fully protective immune responses in neonates after a single vaccination only. Live vaccines generate effective immune responses but have been associated with a number of safety concerns, including improper attenuation and reverting back to virulence.   Nonliving vaccine antigens i.e. whole, inactivated viruses and specifically recombinant or highly purified subunit vaccines are often weakly immunogenic and need adjuvants to enhance the immunogenicity of vaccine based on antibodies and effector T cell functions to prevent infection. Adjuvants are critical components of vaccines, usually used to stimulate faster, stronger, and long-lasting immune responses to vaccines. It has various roles in vaccine formulations i.e. enhance immune responses to vaccine antigens, provide faster onset of immunity, improving immune responses to immunization in infant or elderly populations, whose immune system are immature or waning, dose sparing, either reducing the quantity of antigen required in the vaccine preparation or reducing the immunization schedule (Coffman, Sher et al. 2010). Adjuvants are absolutely required in subunit vaccines due to the poor immunogenicity of such antigens. There is no single universal adjuvant which can cover all the vaccine requirements. Using single adjuvant has a number of limitations, including induction of weak, improper, and short lived immune responses. For example, alum and MF59 (Kenney and Edelman 2003) are the universally approved adjuvant for human vaccines. Both normally induce a Th2 biased immune responses but not cellular immune responses which required for immunity against intracellular infections. In many novel adjuvant technologies, using multiple adjuvants in combination often act synergistically by stimulating and activating a various type of immune cells. Combination of adjuvants platform is promising and beneficial for suboptimal vaccines and particularly advantageous for vaccines against specific and more susceptible populations, such as neonates and the older adults. In some aspects, the neonate immune system is similar to that of the elderly, both having diminished anti-microbial activity by immune cells, reduce d antigen uptake and presentation by antigen presenting cells and compromised adaptive immune responses (Simon, Hollander et al. 2015). Strong adjuvants may provide an approach to boost immune responses in the both neonates and elderly populations. Most of the studies have combined a delivery system with an immunostimulatory adjuvant, especially combinations with the TLR agonists and MPL. Combination adjuvants have only recently been vigorously explored. Some combinations have been tested in humans and large animals and have yielded promising results. Recently, we developed a novel combination adjuvant platform (TriAdj) which is highly effective with wide range of animal and human vaccine. Novel Combination adjuvant platform for animal vaccines: Veterinary vaccines have been used for several of years, and have an important role in protecting animal health, animal welfare, food production, and public health. They are a cost-effective method to prevent animal disease, improve the food production, greatly reducing the need for antibiotics to treat food and companion animals and reduce or prevent transmission of zoonotic disease to human. Interestingly, the challenges related with vaccinating animals are stability, low cost, ease of administration etc., require new solutions in vaccine development. Freund introduced a combination of mineral oils and bacterial cell components (Freunds complete adjuvant) for the improvement of vaccine immune responses (Freund et al., 1937). However, many nations are not using the Freunds adjuvant in animals due to its reactivity and side effects. Alum and emulsions based formulations have been successfully used since long time in a wide variety of animal vaccines. However, with an advanced underst anding of the immune system, many new adjuvants (Saponins, Liposomes, virosomes, particle based and TRL ligand) have recently been developed for veterinary applications. A wide range of adjuvants has been successfully used in commercial vaccines for animals and several new technologies are currently in preclinical development. Over the past decade, we have seen several new combination adjuvants, typically contain two and three individual adjuvant components, including MF59â„ ¢ (Novartis Inc.), ASâ„ ¢ (Glaxo Smith Kline Inc.), IC31â„ ¢ (Valneva Inc.) etc. We recently developed a novel combination adjuvant platform (TriAdj) that is contained of three components, namely toll-like receptor (TLR) agonist either PolyI:C or CpG ODN and an immunostimulatory host defense peptide (HDPs) in polyphosphazene carrier system (Kindrachuk, Jenssen et al. 2009; Garg, Latimer et al. 2013). Synthetic PolyI:C and CpG ODNs are well known potent adjuvant with various vaccine antigens and have been shown to enhance immune responses by activating monocytes/macrophages, dendritic cells, natural killer cells and B cells, and induce the production of proinflammatory cytokines   (Krieg 2002; Trumpfheller, Longhi et al. 2012). The second component, HDPs are derivatives of natural host defense peptides, which are cationic amp hipathic peptides with microbicidal, chemotactic and/or immunomodulatory properties (Yeung, Gellatly et al. 2011). HDPs are involved in a range of immune functions including immune cell recruitment (neutrophils, monocytes, macrophages, T cells and mast cells), innate immune activation, and wound healing (Jenssen, Hamill et al. 2006). The third component, polyphosphazenes are synthetic water-soluble biodegradable polymer with immunostimulatory properties, and forms non-covalent complexes with variety of viral and bacterial antigens and/or other adjuvants to enhance their stability, immunogenicity and allow multimeric presentation (Mutwiri, Benjamin et al. 2007; Andrianov, DeCollibus et al. 2009; Kovacs-Nolan, Latimer et al. 2009; Awate, Wilson et al. 2012). When vaccine antigens were co-formulated with this combination, we found much faster onset of immunity, highly effective even after a single immunization and significantly long lasting, robust, protective immune responses against variety of animal pathogens including bovine viral diarrhea virus, bovine respiratory syncytial virus and porcine epidemic diarrhea virus. The combination adjuvant is stable, cost effective and highly effective in a variety of animals including pigs, sheep, cattle, koalas, cotton rats and mice (Polewicz, Gracia et al. 2011; Khan, Waugh et al. 2014; Snider, Garg et al. 2014; Garg, Latimer et al. 2015). For example, formulation of E2 protein of bovine viral diarrhea virus with TriAdj resulted in strong humoral and cell mediated immune responses, leading to significant protection following pathogen challenge of calves (Snider, Garg et al. 2014). Fusion protein of bovine respiratory syncytial virus co-formulated with TriAdj developed significantly higher antibod ies and interferon gamma secretion (Kovacs-Nolan, Mapletoft et al. 2009). Similarly, formulation of S1 domain of porcine epidemic diarrhea virus, and outer membrane protein of chlamydial major   with TriAdj enhanced humoral and cell mediated immune response in pig and koalas respectively (Khan, Waugh et al. 2014; Makadiya, Brownlie et al. 2016). Novel Combination adjuvant platform for human vaccines: Several combination adjuvants consisting of a variety of immunomodulators such as Immune stimulating complexes (ISCOMs), montanides, nanoemulsions, and Adjuvant Systems have been developed in recent years and are currently being tested with human vaccines in preclinical and clinical trials. The novel adjuvant platform, TriAdj was co-formulated with various human vaccines and tested in mice, pigs, sheep, koalas, and fish. This adjuvant platform is highly effective against a variety of infectious diseases. TriAdj was shown to promote the induction of strong immune responses to various viral and bacterial antigens in multiple animals. For instance, TriAdj in combination with fusion protein of human respiratory syncytial virus (hRSV), mediated the induction of robust, balanced and long-term protective immunity by stimulating long-lived neutralizing antibodies, memory B and CD8+ T cells against hRSV (Garg, Latimer et al. 2013; Garg, Latimer et al. 2014). In addition, mucosal vaccination with TriAdj formulated antigen induced both systemic and local immunity in neonates, even in the face of maternal antibodies (Garg, Latimer et al. 2015). Similarly, when TriAdj was used with pertussis toxoid of Bordetella pertussis, strong and protective immune responses were found in both mice and pigs against lethal infection with B. pertussis (Gracia, Polewicz et al. 2011; Polewicz, Gracia et al. 2011). Furthermore, the vaccine formulated with TriAdj induced a prompt onset, longer duration than existing commercial vaccines and effective after a single vaccination even in the presence of maternal antibodies. (Polewicz, Gracia et al. 2013). TriAdj platform was also formulated with influenza virus antigens or chlamydia antigens, which induces strong immune responses in vaccinated animals (Kindrachuk, Jenssen et al. 2009; Shim, Ko et al. 2010). The combination of adjuvants was also shown to be suitable for maternal immunization. Vaccination of pregnant animals with TriAdj formulated human vaccines resulted in efficient transfer of maternal antibodies and protection from subsequent challenge of the offspring (Elahi, Buchanan et al. 2006; Garg, Latimer et al. 2016). These results indicate that maternal immunization with TriAdj formulated antigens might be an alternative, safe and effective approach to provide protection against pathogens in newborn and young infants. Furthermore, the adjuvant platform can be formulated into microspheres (100 nm to 2 ÃŽ ¼m) to enhance the mucosal and syt emic immune response following intranasal vaccination (Garlapati, Garg et al. 2012). The TriAdj is expected to have multiple applications for the development of vaccines against multiple respiratory pathogens and possibly other infectious agents. Mechanisms of action of novel Combination adjuvant platform: A number of mechanisms of action were identified for this novel combination adjuvant. For example, TriAdj as a mucosal adjuvant increased antigen uptake by dendritic cells, improved dendritic cell maturation, and more efficient transported to local draining lymph nodes to present the antigen to T cells   (Garg, Latimer et al. 2013). TriAdj with antigens promoted the production of chemokines, cytokines and inflammatory cytokines, followed by recruitment and activation of several immune cell populations including dendritic cells, macrophages and neutrophils to the upper and lower respiratory tract, that leads to strong and long-term   protective immune responses of this novel adjuvant formulation (Sarkar, Garg et al. 2016). This was further correlated to the induction of local humoral and cell-mediated immune responses, including production of large numbers of IgA secreting memory B cells as well as effective memory CD8+ T cells. Andrew Marvell’s poem ‘To His Coy Mistress’ Essay

In this essay I will compare and contrast Andrew Marvell’s poem, ‘To His Coy Mistress’, with Elizabeth Barrett Browning’s sonnet, ‘How Do I Love Thee†¦?’ Andrew Marvell’s poem is about an older man trying persuade a younger women to ‘carpe diem’ (seize the day), in order to make love to her, by using compliments and flattery, ‘Vaster than empires, and should go to praise.'(Stanza 1, line 12) Additionally, Elizabeth Barrett Browning’s sonnet is about a female who is expressing her feelings towards a male. Judging by the poem, the woman is deeply in love with the man in a spiritual sense, ‘I love thee to the depth and breadth and height, my soul can reach.’ (Line 2-3) ‘To His Coy Mistress’ is a comparatively long poem of 46 lines, which is divided into three stanzas, representing different parts of the argument for which he is trying to persuade her to sleep with him. In the first stanza, the man flatters the women by using grandiose imagery and hyperbole. He says that her ‘coyness’ would be of no consequence ‘had we but world enough and time’ (Line 1) and then follows with more detail in the following stanzas. The older man also shows how interested he is by expressing the magnitude of his feelings, by explaining how he would, ‘love you ten years before the flood’ (Line 8), even if his love were to remain unrequited, ’till the conversation of the Jews.'(Line 10) In addition he then describes how long he would be prepared to appreciate all of her physical attributes, ‘two hundred to adore each breast, But thirty thousand to the rest.'(Line 15-16) On one hand, he is trying to prove to the young women how much he wants her; on the other hand she could perceive his words in the wrong way, maybe he just wants to sleep with her? You could argue that this is satirising the king’s court, because even though he is flattering her, it is inappropriate to assume that he would be allowed to look at her for this long period of time. In the second stanza, he continues to us the grandiose imagery from the first stanza, but introduces a sense of urgency. When he says, ‘but’ in the very first line, the audience realises that there is going to be a shift in focus. He now says that he hears ‘times winged chariot hurrying near.'(Line 22) From this point, his imagery becomes increasingly desperate. He tries to shock the women into sleeping with him, by talking about her ‘coyness’, and if she perseveres in life that way, she runs the risk of only ‘worms†¦trying that long preserved virginity.'(Line 27-28) He says that his attitude will turn to dust and ‘into ashes all my lust.'(Line 30) This sentence makes the audience more aware of his actual intentions by saying the word ‘lust’ rather than ‘love’, making us believe that he has betrayed his true motives, and slipped up in front of the woman, he is making out he ‘loves’. In the last stanza of ‘To His Coy Mistress’, it sees him almost demand that they make ‘sport’ (love). The imagery on this stanza becomes more erotic, and may have more than one interpretation. ‘Let us roll†¦.up into one ball, and tear our pleasures†¦through the iron gates of life.'(Line 41 and 44) Rolling up into a ball could represent two people making love together or a cannon ball which will smash down, ‘the iron gates of life.’ These gates could represent the woman’s chastity belt or society’s conventions which would frown upon an extramarital affair. The last two lines are similarly ambiguous. They both tie in with lines from the second stanza. ‘Thus, through we cannot make our sun, stand still, yet we will make him run’ (Line 45 – 46), suggests that they should make the most of their time they are spending together, and conceive a child (son). Elizabeth Barrett Browning’s poem, ‘How do I love thee†¦Ã¢â‚¬â„¢ presents quite a contrast to Andrew Marvell’s poem. Its imagery is humble, and very personal. It is an expression of deep love and devotion from one person to another. The purpose of the poem is to quantify the dimensions of her love and at the beginning of the poem, it is very clear to the audience that this woman is deeply in love with her partner By starting with the line, ‘How do I love thee? Let me count the ways'(Line 1) she begins to compare her love to religion and emotions rather than physical attributes, ‘I love thee freely, as men strive for right.’ (Line 7) The imagery she uses to supposedly quantify this love is suggestive of infinity, and a love which has no boundaries, even after the soul is free from the physical body, ‘I shall but love thee after death.’ (Line 14) Constructive descriptions are always used in this poem, compared to Andrew Marvell’s, which also describes the negative issues. Besides Browning’s poem having a very romantic meaning, you could question why the woman needs to prove her love for her partner, because she continually mentions and repeats, ‘I love thee†¦Ã¢â‚¬â„¢ in most of her sentences. She could be indeed counting the ways she loves her partner, however maybe her partner needs reassuring of her love for him. One could argue that, Barrett Browning chose the sonnet form for this poem, for the purposes of contrast; to take something which is supposedly infinitive (her love) and place it in a finite and restricted form (sonnet). Alternatively, people could think different because there are such strict rules governing sonnet writing, (i.e. 10 syllables per line, 14 lines etc) and people might question why she chose such a rigid format, for something which she feels most strongly about. Overall, the two poems are opposites, but they are both concerned with the concept of time, human life and love. At the end of the poem, browning says, ‘if god choose, I shall love thee better after death,'(Line 13-14) showing that even after she dies, her love for this man will continue to grow, loving him for eternity. ‘To His Coy Mistress’, and ‘How Do I Love Thee†¦Ã¢â‚¬â„¢ have many correlations with each other. For example; Both poems are about love; but represent different kinds of love. Andrew Marvell’s poem is about lust and sexual gratification, while Elizabeth Barrett Browning’s poem is about true love and loyalty. The sonnet is written from a women’s point of view, where she expresses her true love for her husband. The other poem is written from a mature mans point of view, and represemts his lust for a younger women. This poem is an elaborate ‘chat up line’ to present a logical argument in order to persuade her to make love to him. This can show how the significance of a poem can differ because of the century it has been written in. ‘To his coy mistress’, was written in the 1600’s while ‘How do I love thee†¦Ã¢â‚¬â„¢ was written in the 1800’s. The cultural difference between these two periods, is the writing style, as in the 1600’s people were interested in composing clever arguments, and were more interested in writing about sex, lust and passion. Whereas in the 1800’s, the poems written had more true meaning, with a deeper, more romantic feeling. ‘And into ashes all my lust’, compared to, ‘How do I love thee†¦Let me count the ways.’ Many love poems are written in a very traditional format, with very strict rules. Elizabeth Barrett Browning took the challenge to write about something which is supposed to have no limits (love) into something which is restricted (sonnet). On the other hand Andrew Marvell’s poem is all based around ‘carpe diem’, in a non-traditional format, with no strict rules or guide-lines. This can affect the imagery used as one has no limitation to the amount of syllables, lines or stanzas, while the other has a restricted format leaving a certain amount of phrases which can be used. Clearly this means that there are different types of imagery used between the two poems since, ‘How do I love thee†¦Ã¢â‚¬â„¢ uses abstract and emotional imagery, which tends to be highly personal and humble. In some cases you cannot quite put a finger on what she is describing. ‘I love thee to the level of every day, most quite nearby sun and candlelight. (Line 5-6) ‘To His Coy Mistress’, tends to use ostentatious hyperbole and grandiose imagery. He uses big overdone, tacky images, by using phrases such as, ‘times winged chariot’, ‘instant fires’ and ‘iron gates of life.’ You can picture what the man is describing; but on the other hand, you can tell that he might be misleading the audience to make them believe that he is in love with this woman. Throughout the whole of my essay, I have come to the conclusion, that both poems have many comparisons, similarities and differences. They both describe a form of love between two people. In my opinion, the poem I prefer is, ‘To his coy mistress’, because it had more of an effect on me, because of the language used. The man comes across as very desperate, but on the other hand with very good charisma. The imagery used is effective, as I could relate and imagine what he is describing. As well as keeping the audience intrigued on what the women will do next, he leaves the ending on a cliff hanger, by not telling on the final decision the women makes! However, I do not dislike the poem, ‘How do I love thee†¦Ã¢â‚¬â„¢ because it has appropriate language for the concept she is describing. I just think it is rather tedious describing only the optimistic qualities about a person, for the reason that everybody has faults and pessimistic attributes. Furthermore, I didn’t find it unique or distinctive from other love poems, since the language and imagery was very similar.

In all the traffic cars the patrol cars have been fitted with specially designed suspension and brakes to cope with the extra load and the varied and demanding conditions they are exposed to. Motorcycle officers also have a specialist role in the policing of major public events, such as football matches, and providing a police escort for VIPs. They co-ordinate roadwork's, speeding complaints and monitor local collision trends. Rapid Response Ambulance Rapid response ambulance is specified to attend emergency incidents, ideally within eight minutes of notification. The Trophy Yellow estate cars, with green reflective Battenburg livery, incident equipment carried on the vehicles includes; oxygen therapy, cardiac defibrillators, entonox, maternity and paediatric pack, burns bag, cervical collars, splints and first aid bag. Plans for major incidents, site maps, body bags, triage cards and contacts for agencies and hospitals are also carried. Additional special items include the light roof bar, radio communications and a reverse-decal ‘Ambulance' livery on the bonnet and ‘RESPONDER' on its tailgate. These cars aren't stationed at the hospital; they are located in areas close to the region boarders making their response time a lot quicker. Their role is to attend emergencies, be first on the scene and aid injured people. Both services are required to attend incidents within a short period of time. To be able to make these deadlines, they are both authorized to use their sirens and lights to make other road users aware that they are responding to an emergency. In many cases both rapid responses are linked together. If theirs an RTA the rapid response would be first on scene that would then get the ambulance service called out if anyone is injured. The rapid response ambulance may take that call if it's within their area, which would attend and wait for ambulance to attend from the hospital to take casualties to hospital. Criminals target vehicles, New cars are automatically fitted with some sort of device attached immobiliser, tracker or alarm. The ideas of these security devices are outlined beneath. Immobilising system, which is automatically activated when the ignition key is removed. The way it works is the device transmits a radio-frequency signal every time the engine is started, sending a different password to the system. Insuring the car cannot be started any other way without the key. Electronic engine immobilisers prevent your car from starting and are the best way to stop thieves. Car alarms can deter thieves from not only stealing your car, but also taking items from it. Older cars are criminals prime targets due to the lack of security protection fitted to them. Steering wheel locks are a cheaper alternative to engine immobilisers use them every time you leave your car. Locking wheel nuts easy to fit and stop thieves from taking your wheels. Have your car registration number or the last seven digits of your Vehicle Identification Number etched onto all windows, both windscreens and your headlamps. Mark all your car equipment, like your car stereo, with your vehicle registration number. Having any of these security measures reduces the attractiveness of the car to thefts. Analysis of driver's attitude whilst travelling on the roads. Due to no one having the same characteristic everyone is different, with his or her own style of driving technique. With this in mind people tend to act differently on the roads. Road rage is the biggest form of driver's attitude on the roads and they aren't necessarily the person behind the wheel either. People speeding, acting irresponsible and general behaviour of others on the roads can cause road rage. Older citizens push the blame onto younger citizens for the way that they drive, younger citizens reverse the blame. There are business people who due to road rage drive more dangerously and faster blaming traffic and deadlines as their excuse. Parents who want to pick children up, but cant park near schools due to heavy congestion around the school. There are people who drive for living truck drivers, van drivers and public transport drivers who are consistently on the roads, that travel through all the peak times of the days. There are the rush hour periods during the day that make everyone less tolerable to waiting calmly. Speeding is mostly influenced by other people speeding, thinking if they can break the limit then so can they, which causes many other problems on the roads. After researching speeding I found that in Northampton there was a work shop for people who had broken the speed limit, where people can analysis there own behaviour on the roads. After the workshop results showed that: 92 per cent of respondents who had been on a Speed Workshop said that they were now more likely to keep to the speed limit. Of those who had received a speeding ticket, but hadn't been on a Speed Workshop, 53 per cent of respondents said that it was not acceptable to exceed the speed limit. The UK legal limit for drivers is 80 mg of alcohol per 100 ml of blood but there is no failsafe guide to the amount of alcohol that a driver can safely consume. The amount and type of alcoholic drink, the weight, sex and metabolism of the driver all play a part. But any amount of alcohol affects driving ability. A motorist's ability to judge speed and distance may be impaired, their reaction times may be slowed and their judgement of risk seriously affected. Advice from the drink-driving site has given this structure for drinking: Immediately before driving: – Men should consume no more than 4 units, women no more than 3 When drinking the night before driving: – Men should consume no more than 10 units, women no more than 7. (This assumes that no alcohol is consumed after 11.30 pm, and that driving does not take place before 8 am the following morning) Cambridge county council have published that in the last three years there have been 324 drink drive accidents involving death and injury. Drink driving may cause the driver not able to judge the speed and distance accurately; the driver may become more overconfident whilst driving endangering other road users. With all these people on the roads, there is no wonder so many people are involved in accidents. Accidents can also be caused by people's personal preferences. A driver may like to drive slower than the person in the car behind due to not feeling confident on the roads or just being cautious. Many drivers on the road don't have enough breaks during travelling that could make the driver tired, irritable and likely to become a danger to themselves and others. Business people travelling in their own car are likely to have mobiles in use, without hands free or microphone the drive by law has to stop where safe and answer the call. As this is unlikely event to happen most people fit attachments in the car to prevent committing an offence. Parents with children should to prevent the driver being distracted, entertain children with some sort of music or equipment that will entertain the children while in their baby seats or whilst they are belted up in the back seats. Using this sort of attitude whilst making a journey will decrease the chance of a common traffic offence and in case of an accident reduce the chance of critical accident. (Ref 1) (Ref 2) (Ref 1) Poster on tiredness – I did a surveyed on ten people asking what they do during long journeys, five people said they don't stop but tend to differ the speed, play music and have sweets in the car. Two people said that they stop every two hours at a pre-selected break while the other three said that it depends on the weather and whether they know where they are going. After showing the poster to the five that don't stop, the drivers have changed their opinion and have agreed to wither stop for a stretch, swap drivers if able to and plan their journey that stop every hour to two hours. (Ref 2) Poster on mobile phones – whilst driving, proves that people aren't multi-skilled that means driving and using a mobile will divide there attention running higher risk of an accident. Surveying the same ten people that I did for the tiredness poster, I asked how many have their mobiles switched on and how they would deal with the driving if they're mobile started ringing. Three people said that they don't have their phone on at all through journeys. Two people said that due to the nature of the business that they have hands free kits and one person said that their company installed a microphone set to avoid missing calls whilst on the road. Four people said that they would only use there mobile whilst sitting in traffic or when they are driving on roads that wont cause the driver distraction. After showing the poster and asking them to read it out, they found it very hard and took a few seconds to work out that the transcript was engaging two different statements. After this experiment, all of the drivers that I surveyed that said that they would at least get some equipment installed in their car to avoid causing accident. Even using hand free kits and microphones aren't advised, but they aren't against the law and they have reduced the amount of accidents. Not everyone can avoid using their mobile whilst travelling due to their job but this does reduce the amount of accidents for the time being.

Here are five sentences that would be much improved by their proper use, or by proper use of other punctuation in cooperation with them. Determine how each sentence would benefit from changes in punctuation and compare your revisions with my suggested solutions at the bottom of the page: 1. â€Å"Not in years, like more than ten years, have I seen someone so committed to owning the stage.† 2. â€Å"Such pioneers trigger and indeed hope for gentrification leading to more and more middle-class home buyers being willing to take a chance on the neighborhood.† 3. â€Å"You, yes you, can say you were there for the advent of the Apple iPod.† 4. â€Å"It’ll take years to know if it works in humans but in mice the tumors almost completely disappeared.† 5. â€Å"Consumer-oriented businesses are trying to find the words, logo, image and, of course, products that will indelibly brand themselves as environmentally friendly.† Answers and Explanations 1. The phrase â€Å"like more than ten years† (with like, as an interjection, separated from the rest of the phrase with a comma), is more emphatic than one that would merely be parenthesized between commas: â€Å"Not in years like, more than ten years have I seen someone so committed to owning the stage.† 2. The clause beginning with leading does not merit being set off from the rest of the sentence with an em dash, but the phrase â€Å"and indeed hope for,† with the interjection indeed bracketed by commas, should be emphasized by being framed by a pair of em dashes: â€Å"Such pioneers trigger and, indeed, hope for gentrification, leading to more and more middle-class home buyers being willing to take a chance on the neighborhood.† 3. â€Å"Yes you,† with a necessary comma between the words, is such an interruptive element that bracketing by a pair of em dashes is necessary: â€Å"You- yes, you- can say you were there for the advent of the Apple iPod.† 4. Just as you’d do in the case of a pair of commas in a sentence that doesn’t sound quite right, diagnostically remove a parenthetical phrase framed by em dashes from an awkward sentence. In this case, â€Å"but in mice† is an essential dependent clause for the second half of the sentence, and the em dash following it is incorrect. The first em dash can be replaced by a comma, or the single dash can be retained: â€Å"It’ll take years to know if it works in humans but in mice, the tumors almost completely disappeared.† 5. Parentheticals are just that interjections, short or long, that are parenthetical to the main sentence, and any parts of speech within them are integral to the interjection alone. Therefore, without the parenthesis set off by em dashes, this sentence lacks a conjunction in the list of three elements preceding the first dash. This means that if someone is willing to do our work than that is acceptable but if he is forced that is unethical so we should not deceive others in doing our will. In compulsion or deceiving others, we violate their will and disrupt their autonomy.The second formulation permits to perform only those actions whose maxim can become a universal law of nature. One acting on maxim requires the test of universalization so that everyone can act on it. It is not necessary that a certain maxim should always be a universal law but it must have to be applied on a universal basis, only that is morally permissible.
